Carisma is expanding into new cell and gene therapy programs and targets and is looking for experienced scientists to drive the leading edge of immunotherapy. Carisma is seeking a Scientist or Senior Scientist to join a growing research team developing a novel immunotherapy for cancer. The team is focused on establishing a lipid nanoparticle (LNP)/mRNA based in vivo CAR-M platform and building a broad oncology pipeline. The technology is being developed by Carisma as part of a recently announced collaboration with Moderna.
The candidate will be responsible for the design and execution of in vivo preclinical studies and will play a vital role in the translation of our novel mRNA-based gene therapies to the clinic. The candidate will be an integral part of a small team, where each member is expected to take responsibility for their actions, work collaboratively and be an ambassador for the company in front of leadership, vendors, partners, and others. The level of the position and compensation package will be competitive and commensurate with experience and the specific level of the candidate.

The position is based in Philadelphia, PA.

Responsibilities

    • Develop novel in-vivo tumor models in a variety of murine systems to support development of novel immunotherapy platforms.
    • Execute and oversee murine biodistribution, efficacy, and translational studies in xenograft, humanized, or syngeneic murine models of cancer.
    • Work with the research team to collaboratively evaluate dynamics in the tumor microenvironment using state of the art techniques such as high parameter flow cytometry, single cell RNA sequencing, and others.
    • Prepare and deliver data presentations to management.
    • Execute a variety of in vivo technical skills including mouse handling, dosing, and bleeding.
    • Process samples ex vivo to obtain readouts including cell infiltration, cell phenotype and cytokine levels
    • Appropriately analyze data, interpret results, and maintain accurate laboratory notebook records; prepare and deliver presentations at team meetings
    • Trouble shoot/optimize experiments and procedures
    • Keep current with advances in the company’s area of research interest
    • Supervise a small team of research associates performing in vivo experiments
    • Flexibly support multiple research projects as the company grows

Requirements

    • PhD in life sciences or related discipline, with a minimum of 3 years’ relevant experience, or MS with 8 years of experience in the field. Cancer immunotherapy expertise is preferred.
    • Demonstrated record in in vivo tumor models, including xenograft and syngeneic tumor models, is essential.
    • Experience with hands-on mouse handling (i.e. tumor progression monitoring, nescropsy tissue collection and processing, bleed collection) is essential.
    • Independent experience with intravenous, intraperitoneal, and subcutaneous injections in mice is required.  
    • Experience with flow cytometry, cell based functional assays, cell line culture, primary immune cell culture, transduction/transfection, and cytokine release (ELISA/MSD) is required.
    • Ability to work both independently, lead a small team and coordinate with other teams to execute large studies is required
    • Must have excellent communication, both oral and written, with the ability to perform detailed dataanalysis, keep accurate records and present.
