Mammoth is seeking a Scientist with expertise in conducting genetic screens in E. coli to join its growing Discovery team. The Discovery group is responsible for developing novel CRISPR-associated (Cas) nucleases for genome editing and diagnostic applications.
The successful candidate will be responsible for executing and developing new E. coli-based genetic screens to characterize novel and engineered Cas nucleases. The scope of the genetic screens will include the determination of (i) PAM specificity, (ii) optimal guide RNA processing sites and (iii) mutation tolerance between guide RNA and target sequence. Over the first year of employment, the successful candidate will function as an individual contributor with efforts split 50-50 between execution and tool development. After the first year, the successful candidate will have the opportunity to re-shape their role to suit their career development goals.


    • Design and execute genetic screens in E. coli
    • Work with the Director to set quarterly and annual goals for E. coli-based genetic screens.
    • Develop Mammoth’s platform (assays, tools and infrastructure) for E. coli-based genetic screens.
    • Document and regularly present experimental results at internal meetings
    • Train and Mentor Research Associates
    • Coordinate with Research Associates and Computational Biology staff to execute experiments


    • 3 years experience conducting genetic screens in E. coli
    • PhD or 5 years experience in the synthetic biology or biotechnology industry

Preferred Qualifications

    • Direct experience characterizing novel Cas effector nucleases
    • Deep familiarity with CRISPR-Cas literature
    • 2 years synthetic biology or biotechnology industry experience with PhD
    • Hands-on experience using E. coli as a host organism for HTP screening in 96 or 384 well plate format
    • Pooled screening experience
    • Hands-on experience using laboratory automation (liquid handlers and colony pickers) to execute experiments
    • Hands-on experience with NGS assay development
    • Computer and scripting skills (Benchling, Geneious, JMP, SQL, R, python)
